Laser desorption/ionization mass spectrometry analysis of small molecules in biological samples

Description:
The project deals with the development of matrix-assisted and matrix-free laser desorption/ionization mass spectrometry (MA)LDI-MS for the analysis of small molecules in biological samples. Samples of rat urine, plasma and tissues will be obtained after selected drug administration together with placebo samples with subsequent drug metabolite analysis. Further, samples of human tissues (health x tumor) will be analyzed with the aim to obtain information about present biomarkers of tumor disease and possible future application in the early clinical diagnostics. Particular samples will be characterized using MALDI imaging to obtain 2D images with the information about present small molecules, their amount and distribution in studied tissues. Parallel measurements using ultra high performance liquid chromatography/atmospheric pressure ionization tandem mass spectrometry (UHPLC/API-MS/MS) will be performed for the pro ling of individual group of compounds with the emphasis on the elemental composition determination based on the high mass accuracy and characteristic fragmentation.
