Determination of lipolytic activity using p-nitrophenylpalmitate as a substrate is based on ability of lipolytic enzymes to hydrolyse this substrate. p-nitrophenol is formed and its absorbance is measured at the wavelength 410 nm. The activity of lipase is determined using calibration curve for p-nitrophenol, i.e. is expressed as amount of p-nitrophenol which was releases from p-nitrophenylpalmitate by enzyme. The activity of enzymes depends on concentration of hydrogen ions (pH) in medium. It is possible to determine the value of pH for which the activity of enzyme is the highest. (pH optimum).
