The project focuses on the extension of the application range of supercritical fluid chromatography hyphenated to mass spectrometry (SFC/MS) for the analysis of polar and ionic metabolites in biological matrix. The optimization of the quantitative sample preparation protocol, the improvement of the chromatographic performance by evaluating various stationary phases and mobile phase compositions for the separation of polar and ionic compounds will be investigated. The optimized method will be validated and applied for the determination of concentration profiles in body fluids of healthy and diseased volunteers. Special focus will be paid to determine metabolic profile differences depending on the sample collection procedures, i.e., the use of serum or plasma blood collection tubes. Multivariate data analysis will be applied for the determination of statistical relevant compounds reflecting a certain sample type. Results will be compared to UHPLC/MS to give evidence that SFC/MS is complementary and suitable for biological studies.
