Description:
The proposed project is focused on the development of a new interface type with active focusing of transferred fractions in comprehensive two-dimensional liquid chromatography. New interface (modulator) will be characterized, which enables automated real-time adjustment of focusing conditions based on monitoring of selected physical-chemical variables of the eluate leaving a first dimension column, or based on monitoring of the focusing process utilizing electromigration principles. New interface will be tested using systems with regular analytical columns and with capillary microcolumns. The microcolumns based on organic monoliths for reversed-phase, and hydrophilic interaction chromatography will be prepared within the project. New procedures of preparation of biological samples will be developed suitable for the following analyses using two-dimensional liquid chromatography with the developed interface. The main outcome of the project will be the methods used for sensitive analysis of biological samples coupled with mass spectrometric detection.
