eng Observation of Antioxidative Activity of Uric Acid in Men Uric acid, the final product of human purine metabolism, can be oxidized by various reactive oxygen species to allantoin. Therefore, the allantoin concentration may correlate with the extent of oxidative stress in humans. Aim: To compare allantoin concentration in plasma and erythrocytes of the patients with oxidative stress and the control group to verify this hypothesis. Concentration of allantoin was measured after its derivatization to glyoxylate-2,4-DNPH in plasma and erythrocytes obtained from patients with chronic renal failure before hemodialysis (n = 30, 15 women and 15 men) and from group of blood donors (n = 30, 15 women and 15 men) using HPLC (reversed phase, with gadient elution and UV-VIS detection at 360 nm). We found the significant differences in allantoin concentration between the patients and the control group both in plasma (20.5 +- 6.5 micoM vs. 2.7 +- 0.7 microM; p < 0.001) and erythrocyres (82.8 +- 39.1 nmol/g Hb vs. 21.8 +- 6.1 nmol/g Hb; p < 0.001). Statistical significance was evaluated by Student's t-tes. P < 0.001 values were concidered statistically significant. Conclusion: Significantly higher levels of allantoin both in plasma and erythrocytes of patients with chronic renal failure indicate that allantoin may be used as a good marker of oxidative stress.
