eng In present work, the applicability of capillary zone electrophoresis coupled with LIF detector as a powerful clinical diagnostic and prognostic tool for certain diseases resulting in the alteration of glycan profiles is demonstrated. Derivatized neutral and acidic N-linked glycans released from human glycoproteins from healthy and breast cancer patients were resolved using a polyacrylamide coated capillary with reduced electroosmotic flow. Glycan structure identification was elucidated using a combination of strategies, including comparison with standard glycan libraries and the enzymatic digestions of N-linked glycan pools using specific and non-specific exoglycosidases. By doing so, 47 glycan structures, many being structural isomers, were identified. Due to its ability to resolve structural isomers, CZE-LIF technique complements other means for glycan mapping such as a mass spectrometry. CZE;N-glycans;Clinical diagnostics;Cancer
