Vitamín B6 (pyridoxin) je obecným názvem pro tři strukturně podobné, biologicky aktivní formy vitamínu B6, pyridoxalu, pyridoxolu a pyridoxaminu. Významným biologicky neaktivním metabolitem, vylučovaným ledvinami, je kyselina 4-pyridoxová. Metody. Vitaméry B6 (pyridoxal 5?-fosfát a pyridoxal) a kyselinu 4-pyridoxovou jsme stanovovali pomocí kapalinové chromatografie s fluorescenční detekcí. Pro zvýšení intenzity fluorescence pyridoxal 5?-fosfátu a pyridoxalu jsme před HPLC analýzou vzorky derivatizovali pomocí semikarbazidu. Kyselina 4-pyridoxová je přirozeně silně fluoreskující látkou. Pro separaci pyridoxal 5?-fosfátu, pyridoxalu a kyseliny pyridoxové jsme jako stacionární fázi vybrali kolonu C18 (125 mm x 4 mm, velikost částic 5 um) a mobilní fází byl 60 mmol/l Na2HPO4 v methanolu (90,5:9,5, v/v). Výsledky. Hladiny pyridoxal 5?-fosfátu, pyridoxalu a kyseliny pyridoxové jsme stanovovali u skupiny dobrovolných dárců krve, pacientů s kardiovaskulárním onemocněním, pacientů s diabetem mellitus a pacientů s metabolickým syndromem. Předběžné referenční hodnoty pyridoxal 5?-fosfátu, pyridoxalu a 4-pyridoxové kyseliny u skupiny dobrovolných dárců krve byly 42,2 +/- 6,8 nmol/l, 3,5 +/- 0,9 nmol/l a 18,0 +/- 2,4 nmol/l. Závěry. Hladiny jednotlivých vitamérů u vybraných skupin pacientů a hladiny u skupiny dobrovolných dárců krve byly porovnávány. Kromě hladin pyridoxalu u pacientů s diabetem mellitus jsme žádné statisticky významné rozdíly nenašli. Poděkování. Vitamin B6 (pyridoxine) is a global term for three structurally related biologically active forms of vitamin B6, which are pyridoxal, pyridoxol and pyridoxamine. Important metabolite excreted in urine is biologically inactive 4-pyridoxic acid. Methods. Determination of vitamers B6 (pyridoxal 5'-phosphate and pyridoxal) and 4-pyridoxic acid has been done by using high-performance liquid chromatography with fluorescence detection. Pre-column derivatization using semicarbazide has been used for increasing intensity of pyridoxal 5'-phosphate and pyridoxal fluorescence. 4-Pyridoxic acid is a naturally strongly fluorescing compound. A column C18 (125 mm x 4 mm, particle size 5 um) and a mobile phase containing 60 mmol/L Na2HPO4 / methanol (90.5:9.5, v/v) has been chosen for separating pyridoxal 5'-phosphate, pyridoxal and 4-pyridoxic acid. Results. This method has been applied for determination of concentration of pyridoxal 5'-phosphate, pyridoxal and 4-pyridoxic acid in a group of blood donors, patients with cardiovascular diseases, patients with diabetes mellitus and patients with metabolic syndrome. The preliminary reference range of pyridoxal 5'-phosphate, pyridoxal and 4-pyridoxic acid in a group of blood donors were 42.2 +/- 6.8 nmol/L, 3.5 +/- 0.9 nmol/L and 18.0 +/- 2.4 nmol/L, respectively. Conclusions. Statistical analysis of measuring data has been done including comparison of values obtained from blood donors to values obtained from patients with diseases. There has not been found statistically significant differences of data except pyridoxal values of patients with diabetes mellitus.
