Cílem práce bylo vyvinout HPLC metodu pro simultánní stanovení ubichinolu-10 a ubichinonu-10 v lidské plazmě. Krev byla odebírána do zkumavek obsahující EDTA. Po centrifugaci byla plazma deproteinována methanolem a extrahována hexanem. Hexanová vrstva byla dávkována na kolonu 125 x 4 mm RP-C18 (Purospher, 5 um; Merck). Složení mobilní fáze bylo následující: methanol-2-propanol-octan sodný. Mobilní fáze byla filtrována přes 0,45-um nylonový filtr a odvzdušněna. Průtok mobilní fáze byl 0,5 ml/min a kolona byla termostatována při 37 °C. Koenzym Q10 byl detekován elektrochemickým detektorem ESA Coulochem II 5100A. Ubichinon-10 byl pomocí kondicionální cely (E = -600 mV) redukován na ubichinol-10. Poté byl ubichinol-10 detekován pomocí analytické cely Model 5010 (E1 = -150 mV, E2 = +600 mV). Jako vnitřní standard jsme používali ubichinon-9. Analytické parametry metody byly vyhovující, variační koeficient přesnosti v sérii i mezi sériemi byl pod 10%. Vyvinuli jsme HPLC metodu pro simultánní stanovení ubichinolu-10 a ubichinonu-10 s dostatečnou citlivostí, správností a linearitou. Meoda je vhodná pro stanovení obou forem koenzymu Q10 v biologických vzorcích.	Koenzym Q10; HPLC

Ubiquinol-10, the reduced form of coenzyme Q10 protects cells from oxidative damage. The oxidized form of coenzyme Q10 is known as ubiqinone-10. The ubiquinol-10/ ubiquinone-10 ratio may be used as a marker for oxidative damage. The aim was to develop a HPLC method for the simultaneous determination of ubiquinol 10 and ubiquinone 10 in human plasma. Plasma for analysis was collected into vacutainer tubes containing EDTA. Tubes were centrifuged and extracted with hexane and methanol. Sample was injected onto 125 x 4 mm RP-C18 column (Purospher, 5 um; Merck). The mobile phase consisted of methanol-2-propanol-sodium acetate. We had filtered this mobile phase through a 0.45 um nylon filter and than deaerated before use. The flow rate was 1 ml/min and temperature was set to 37°C. We detected coenzyme Q10 with ESA Coulochem II 5100A electrochemical detector. After separation, the components passed the conditioning cell (-600 mV) and ubiquinone-10 was reduced to ubiquinol-10. We detected ubiquinol-10 using Model 5010 analytical cell and with electrode 1 set at screening potential of -150 mV and electrode 2 set at +600 mV. We used ubiquinone-9 as internal standard. Analytical performance of this method is satisfactory, both intra-assay and inter-assay were with coefficient of variation below 10%. We have developed a HPLC method for the simultaneous determination of ubiquinol 10 and ubiquinone 10, which showed satisfactory analytical sensitivity, precision and linearity.
