eng Identification of Czech Arcobacter isolates using multiplex PCR, 16S rDNA and 16S/23S rDNA intergenic spacer sequence analysis. 16 isolates of G- aerotolerant, spiral shaped bacteria isolated from different matrix (fresch chicken meat, waste water from meat concerting plants, environmental sample from poultry farm in Czech Republic) were identificated by multiplex PCR assay. 15 strains examinated bacteria were confirmed as Arcobacter butzleri, 1 strain was confirmed as Arcobacter cryaerophilus. These isolates were further identified by the sequencing of 16S rDNA and intergenic region between 16S and 23S rDNA genes. Bacterial DNA was isolated using GenElute Bacterial DNA Kit (Sigma). The 16S rDNA and intergenic spacer was amplified by PCR using universal bacterial primers. The PCR product was isolated from agarose gel by TaKaRa RECOCHIP (TaKaRa, Japan), consequently cleaned using PCR ClenUp Kit (Pure Biotech, USA) and directly sequenced. The dye terminator sequencing was used for all sequencing analyses with ABI DT v. 3.1 Kit (Applied Biosystems, USA) on the ABI-3100 Avant four-capillary DNA sequencer (Applied Biosystems, USA). Sequences of Czech isolates of Arcobacters were compared with sequences from type strains of Arcobacter butzleri CCUG 30484, Arcobacter cryaerophilus ATCC 43157, Arcobacter cryaerophilus ATCC 43158, Arcobacter skirrowii CCUG 10375 and by the public DNA database queries. arcobacter; multiplex PCR;16S rRNA;16S rDNA; intergenic spacer
