Aflatoxins are potent carcinogenic, mutagenic and teratogenic metabolites produced predominantly by A. flavus, A. parasiticus, A. nomius and A. tamarii. PCR methods are frequently used for detection of pathogenic or toxinogenic microorganisms. For detection, unique sequences of these microorganisms are chosen targets, which are predominantly toxin or pathogenicity genes. The first step fungi determination is DNA isolation. The most difficult step in the isolation of DNA from Aspergillus species is to disrupt cell wall without causing damage to genomic DNA. Classical techniques of DNA extraction were optimized and compared with use of commercial PCR kits. These methods were classified in view of DNA purity, concentration, speed of isolation, intensity of instrument techniques and manipulation and were taken single isolation cost into consideration. Methods were optimized by using pure cultures A. parasiticus var.globosus CCM F-550 and A. flavus CCM F-108. Obtained DNA was amplified by PCR. Specific PCR product was detected by electrophoresis. Records were interpreted. Specific PCR product will be use for electrochemical detection. aflatoxins,DNA, isolation
