eng Optimization of Selectivity, Peak Capacity and Separation Time in Two-Dimensional HPLC by Matching Separation Systems and Adjusting Gradient Conditions in the Two Dimensions The main objectives of two-dimensional (2D) LC-LC separation techniques are: 1/ significant increase in the peak capacity, i.e., in the number of sample compounds separated in a single run in complex samples with respect to single-dimensional separations, and 2/ possibilities of sample separation into distinct classes on the basis of structural similarities. In-line "comprehensive" LCxLC has experienced rapid development during the past years, mainly because it allows automatic performance and significantly increased number of compounds separated per the time unit in comparison to single-dimension HPLC separations. The operation conditions in the two dimensions should match to provide maximum twodimensional peak capacity in the time available for the analysis. The chemistry of the stationary phase and the composition of the mobile phase should be optimized to provide noncorrelated retention in the two dimensioal "orthogonal" LCxLC separation systems.
