cze Stanovení kreatininu v lidském séru metodou HPLC s UV detekcí: porovnání s komerční enzymatickou a rutinní spektrofotometrickou metodou Je zde popsáno citlivé a specifické stanovení kreatininu v lidském séru. Výsledky u 232 vzorků lidských sér získané metodou HPLC byly porovnány s enzymatickou a spektrofotometrickou (Jaffe) metodou. Vzorky sér byly deproteinovány ethanolem a analyzovány metodou RP-HPLC. Pro separaci byla vybrána analytická kolona MAG 1, 4,6 x 250 mm, Labiospher PSI 100 C18, 5 um. Mobilní fází byla směs 3% ethanolu v 25 mmol/l NaH2PO4 (v/v), pH 6,50. Analytické parametry metody byly vyhovující. Výsledky získané metodou HPLC dobře korelovaly jak s enzymatickou, tak také se spektrofotometrickou metodou. Spektrofotometrická metoda vykazovala téměř podobné výsledky jako HPLC metoda. Metodou HPLC byly získány nejnižší hodnoty u pacientů s non-insulin dependentním diabetem mellitus a u pacientů se zvýšenou hladinou bilirubinu. To poukazuje na to, že při Jaffe metodě reaguje s pikrátem mnoho interferujících látek, jako například glukosa a bilirubin. kreatinin; lidské sérum; HPLC; porovnání metod
eng Determination of creatinine in human serum using HPLC with UV detection: A comparison with commercial enzymatic and routine spectrophotometric methods A sensitive method for the specific determination of creatinine in human serum with high precision and accuracy is described. results from measurement by high-performance liquid chromatography on 232 serum samples were compared with enzymatic and spectrophotometric (Jaffe)methods. Serum samples wre deproteinized with ethanol and analyzed by RP-HPLC. For the separation, a reversed phase column MAG 1, 4.6 x 250 mm, Labiospher PSI 100 C18, 5 um was used. The mobile phase consisted of 3% ethanol in 25 mmol/L NaH2PO4 (v/v), pH 6.50. The analytical performance of this method was satisfactory. Results obtained by chromatographic method correlated with an enzymatic method, as well Jaffe's method. Spectrophotometric method gave almost similar values as HPLC method. The lowest values at patients with non-insulin dependent diabetes mellitus and with increased bilirubin level were obtained by HPLC. It could be caused by non-specific interferents in the case of Jaffe's method. creatinine; human serum; HPLC; methods comparison
