eng Lipidomic Profiling of Biological Tissues and Plasma Using Off-line 2D-LC-MS/MS Lipidomics contributes towards the understanding how lipids function in a biological system and for the elucidation of the mechanism of lipid-related diseases.For this purpose, complex lipidomic approach using off-line two-dimensional HPLC/MS/MS is developed together with the complementary information from conventional gas chromatography - flame ionization detection profiling of fatty acids after the transesterification of lipid extracts. The total lipid extract was fractionated into individual lipid classes using hydrophilic interaction liquid chromatography (HILIC) in the first dimension. The isolated fractions of individual lipid classes are separated using reversed-phase HPLC with C18 porous shell particles column in the second dimension. Our optimized off-line 2D-LC/MS/MS method has been applied for various plant, animal and human tissues and plasma samples. The retention behavior and important fragment ions observed in mass spectra and their use for the identification of lipids are described. Lipidomics; plasma; LC/MS; GC/FID; HILIC
