eng Optimization of lipidomic analysis using off-line two-dimensional HILIC x RP HPLC/MS Lipids are important components in all biological tissues having many essential functions associated with the proper function of organisms. The disruption of lipid metabolism can lead to serious diseases. For this purpose, lipidomic analysis using off-line two-dimensional coupling of hydrophilic interaction liquid chromatography (HILIC) and reversed phase (RP) high-performance liquid chromatography mode with mass spectrometry detection was optimized. In the first dimension, total liquid extract was fractioned using HILIC into individual lipid classes. Optimized HILIC separation enables the fractionation of 18 lipid classes that cover a wide range of polarities. Collected fractions were separated using RP method into individual lipid species.Individual lipid species and their fatty acid composition and position of fatty acyls on the glycerol skeleton was identified using mass spectrometry. Off-line coupling of HILIC and RP-HPLC modes enables the comprehensive analysis of plant and animal samples.
