eng Separation of Triacylglycerol Enantiomers and Regioisomers using Chiral HPLC/MS In this work, method for the analysis of regioisomers and optical isomers in one run have been developed based on the separation of triacylglycerols using chiral stationary phase in the normal-phase mode. In this system, the retention behavior of triacylglycerols is governed by the number of double bonds. Separation of triacylglycerol enantiomers and regioisomers using optimized HPLC conditions is demonstrated on the analysis of mixtures of triacylglycerol standards prepared from selected mono-acyl triacylglycerols (R1R1R1) containing fatty acids commonly presented in the natural samples by the randomization reaction providing mixtures of all isomers. Triacylglycerol regioisomers are identified based on different ratios of fragment ions formed by losses of fatty acids from the glycerol skeleton in APCI mass spectra, while optically pure triacylglycerol standards are prepared for the identification of retention order of enantiomers. triacylglycerols;chiral chromatography;HPLC
