eng Estimation of glutathione reductase inhibition in presence of glutathione depleting agents Glutathione reductase (GR) is a key enzyme of glutathione metabolism. This enzyme catalyzes NADPH-dependent reduction of glutathione disulfide (GSSG) to reduced form (GSH). The main goal of our study was to find out if potent glutathione depleting agents possess also an inhibitory effect in glutathione reductase. We measured activity of glutathione reductase by spectrophotometric method in presence of glutathione disulfide (8 mM) and NADPH (0.8 mM). The decline of absorbance was determined at 340 nm (Tecan Infinite M200). We used purified yeast glutathione reductase (Saccharomyces cerevisiae, 100 mU/ml). We tested a variety of concentrations of ethacrynic acid (0 - 1000 μM) and diethyl maleate (10 mM). We conclude that model glutathione depleting agent – ethacrynic acid inhibites glutathione reductase activity additionally to glutathione depletion. On the other hand, a toxic effect of diethyl maleate is caused strictly by glutathione depletion.
