eng Sperm fatty acid composition in men The fatty acid composition of spermatozoa plays an important role for successful fertilization. The lipids are important for the membrane fluidity and flexibility. Lipids, mainly polyunsatured fatty acids, are susceptible to peroxidation, resulting in functional disorder of the sperm. Therefore lipoperoxidation may lead to men infertility. The aim of this study was to investigate the saturated and unsaturated fatty acids composition of phospholipids of spermatozoa. We have studied the phospholipid fatty acids in men spermatozoa. Semen samples were obtained from consecutive male partners of couples presenting for a fertility evaluation at Sanus, In Vitro Fertilization Clinic, Pardubice. After liquefaction at room temperature the raw semen specimens were divided into two portions, the first to estimate the sperm concentration, sperm morphology and sperm motility (according to the WHO guidelines, 2010), the second to be stored at -80°C. After thawing at room temperature, the raw semen specimens were centrifuged (10 min., 4°C, 22000g), supernatant was carefully removed and the pellet was used further. After protein precipitation, lipids were extracted and separated by preparative thin-layer chromatography into five classes (phospholipids, diacylglycerols, free fatty acids, triacylglycerols and cholesterol esters). Phospholipid fraction was isolated and after the addition of an internal standard (cis-13,16,19-docosatrienoic acid) was hydrolyzed and converted to methyl esters of fatty acids. This way prepared samples were analyzed by gas chromatography. polyunsaturated fatty acid; saturated fatty acid; sperm; gas chromatography
