cze Frakcionace tokem v asymetrickém tokovém poli Kapitola podává přehled teorie a principů separace tokem v asymetrickém tokovém poli (A4F). Je diskutován vliv různých operačních parametrů na rozlišení v A4F. Pro lepší pochopení metod A4F je porovnávána s rozměrově vylučovací chromatografií (SEC). Na rozdíl od SEC, kde hlavní separační mechanismus spočívá v rozměrově omezené permeaci makromolekul do pórů různé velikosti, A4F separuje molekuly bez příspěvku stacionární fáze. To je nejvíce významný rys této techniky, který přináší několik výhod oproti SEC. Pozornost je věnována kombinaci A4F s víceúhlovým detektorem rozptylu světla (MALS), který umožňuje stanovení distribuce molárních hmotností, velikosti a charakterizaci větvení. eng Asymmetric Flow Field Flow Fractionation The chapter provides overview of the theory and principles of asymmetric flow field flow fractionation (A4F). Effect of various operational parameters on the resolution of the A4F technique is discussed. For the sake of better understanding the A4F method is contrasted with widely known size exclusion chromatography (SEC). In contrast to SEC, in which the size restricted permeation of macromolecules into the pores of different size is the major separation mechanism, A4F separates the molecules without contribution of stationary phase. That is the most distinguished feature of the method that offers several advantages over SEC. The focus is on the combination of A4F with a multi-angle light scattering (MALS) detector that allows direct determination of molar mass and size distribution and characterization of branching.
