Infusions Using Micellar Electrokinetic Capillary Chromatography and Liquid Chromatography-Tandem Mass Spectrometry Inflorescence of Sambucus nigra L. is used in the form of water infusions to treat the symptoms of influenza, colds and sinusitis due to their diaphoretic properties [1]. The biological effects can be attributed to the presence of plant metabolites, which are mainly (poly)phenolic compounds. In the present work, we have developed and compared two methods for the determination of the content of (poly)phenols in Sambucus nigra L. infusions. The liquid chromatographic method based on reversed-phase separations using octadecyl silica gel column packed with fused-core particles was employed in combination with the electrospray ionization tandem mass spectrometric detection n the negative-ion mode. The individual compounds were identified based on the multiple reactions monitoring of characteristic transitions in the mass spectra. The second micellar electrokinetic capillary chromatography method was employed using non-coated fused silica capillary and sodium decyl sulfate micellar agent, which offers increased selectivity and speed of the analysis for the separation of (poly)phenols in comparison to the most commonly used sodium dodecyl sulfate [2]. The parameters of the quantitative analysis and the results were compared using analysis of variance and the effect of the sample preparation on the content of (poly)phenols in water infusion was evaluated. The results for both methods were correlated and the differences are discussed with the aim to provide a tool for the comprehensive characterization of the herbal infusion samples.
