Rozšířili jsme velikost a podíl mezopórů pomocí polymerace relativně polárního delšího řetězce síťovadla s lauryl-methakrylátovým monomerem pro přípravu monolitických kolon pro separaci a RP s obojetným sulfobetainem pro aplikace v HILIC. Kolony ukazují dvojí zádržný mechanismus, HILIC ve vodně-organické mobilní fáze s vysokou koncentrací acetonitrilu a RP ve vodném prostředí. Off-line dvojrozměrná LC separace může být provedena nejprve s rostoucí (režim RP), a poté se snižující se koncentrací (HILIC) acetonitrilu ve vodném pufru. Následné HILICxRP a RPxRP dvourozměrné separace může být dosaženo pomocí jediné obojetné monolitické kolony v první dimenzi. Tyto HILICxRP a RPxRP chromatogramy byly spojeny k vytvoření trojrozměrného LC systému. Organic polymer monolithic columns provide excellent rapid gradient separations of proteins and other biopolymers; however they usually show too low efficiency for separation of small molecules, because of low proportion of mesopores. Continuing attempts at improving the pore morphology were reported by adjusting the time, temperature and porogen solvents used in the polymerization reaction, or by post-polymerization modification of the polymer surface. We increased the size and proportion of mesopores by using polymerization of relatively polar longer-chain cross-linkers with lauryl methacrylate monomer to prepare monolithic columns for RP separations and with a zwitterionic sulfobetaine monomer for HILIC applications. The columns show dual retention mechanism, HILIC in aqueous-organic mobile phases with high concentrations of acetonitrile, and RP in more water-rich environment. Off-line two-dimensional LC separations can be accomplished in alternating runs on a polymer zwitterionic polymethacrylate monolithic column, first with increasing (RP mode) and then with decreasing (HILIC mode) gradients of acetonitrile in aqueous buffers. The separations in the off-line two-dimensional system on a single column showed high orthogonality and excellent reproducibility. A zwitterionic polymethacrylate monolithic 0.5 mm i.d. micro-column can be used in the first dimension to be coupled on-line with a short column in the second dimension, where either a core-shell, or a silica-based monolithic non-polar column can be used in a comprehensive LCxLC setup. Subsequent HILICxRP and RPxRP two-dimensional separations can be achieved with a single zwitterionic monolithic column in the first dimension. The HILICxRP and RPxRP chromatograms were combined to provide a three-dimensional LC systém.
