cze Ultracentrifugace krevní plazmy Úvod: Na základě rozdílné hustoty lze lipoproteiny dělit pomocí ultracentrifugace na HDL (High Density Lipoprotein), LDL (Low Density Lipoprotein) a VLDL (Very Low Density Lipoprotein). Každou vrstvu charakterizuje jiná proteinová složka lipoproteinu, tzv. apoprotein (apo-A, apo-B100 a apo-c). Lipoproteiny jsou významným rizikovým faktorem pro řadu onemocnění, a proto je velmi důležité sledovat jejich koncentraci v již zmíněných frakcích. Metoda: Pro vakuovou ultracentrifugaci byl použit přístroj OptimaTM Preparative Ultracentrifuge, Palo-Alto, USA. Plazma i její frakce, které byly ultracentrifugovány, byly analyzovány pomocí biochemického analyzátoru ADVIA 1800. Výsledky: Bylo zjištěno, že rozdělení plazmy na frakce HDL, LDL a VLDL v jednom kroku není možné, a proto byla použita následná ultracentrifugace. Pro následné oddělení frakcí byly použity dělící roztoky o hustotách ρ = 1,006 g.cm-3, 1,063 g.cm-3 a 1,117 g.cm-3. Bylo optimalizováno i množství plazmy pro ultracentrifugaci v závislosti na použitém rotoru. Závěr: Plazmu lze velmi dobře rozdělit pomocí ultracentrifugace na tři základní frakce, které byly charakterizovány pomocí apoproteinu apo-A a apo-B100. Nejlepší rozdělení (96 - 105 %) bylo dosaženo s výchozím množstvím plazmy 1,5 ml. ultracentrifugace, lipoproteiny, HDL, LDL, VLDL
eng Ultracentrifugation of plasma Introduction: On the basis of differing density lipoproteins can be separated via ultracentrifugation HDL (high density lipoprotein), LDL (Low Density Lipoprotein) and VLDL (Very Low Density Lipoprotein). Each layer is characterized by different protein component of lipoproteins, called. apoprotein (apo-A-apo B100 and apo-c). Lipoproteins are a major risk factor for many diseases, and therefore it is very important to monitor their concentration in the mentioned fractions. Method: For ultracentrifugation vacuum device was used optimates Preparative Ultracentrifuge, Palo-Alto, USA. Plasma and its fractions which were ultracentrifuged were analyzed by biochemical ADVIA 1800's. Results: It was found that separation of plasma fractionating HDL, LDL and VLDL in one step is not possible, and therefore was used subsequent ultracentrifugation. For the subsequent separation of the fractions was used dividing solutions with densities ρ = 1.006 g.cm-3, 1.063 g.cm-3 and 1.117 g.cm-3. Was also optimized for the amount of plasma ultracentrifugation depending on the rotor. Conclusion: The plasma can be easily separated by ultracentrifugation on three basic fractions were characterized by apoprotein apo A and apo B100. best division (96 – 105 %) was achieved with the initial quantity of 1.5 ml plasma. ultracentrifugation, lipoprotein, HDL, LDL, VLDL
