eng Orthogonality of Liquid Chromatography and Capillary Electrophoresis in Two-dimensional Separation of Oligosaccharides The analysis of biologically important carbohydrates remains a challenge due to their high structural complexity and the current method development is usually focused on the simplification of sample preparation, new separation approaches and mass spectrometry applications. For the chemical substances with diverse structures and functional groups as the mixtures of glycans, the peak capacity necessary for their successful separation is significantly higher, than the value achievable by uni-dimensional separations. One of the promising approaches for the analysis of biological compounds is the combination of liquid chromatography and capillary electrophoresis in two-dimensional system. In the present work, we have investigated the orthogonality of various liquid chromatographic phase systems including reversed-phase and hydrophilic interaction liquid chromatography with the capillary electrophoretic separation of oligosaccharides. To speed-up the CE separation, we have employed capillaries with covalently bonded neutral polymers such as polyvinyl alcohol, polyacryl amide and polyethylene glycol. The different derivatization procedures of oligosaccharides were compared based on the reductive amination reaction. Finally, the dimensions of the liquid chromatographic columns, the flow rates and the dimensions of the electrophoretic capillaries were optimized for the application of both techniques in on-line two-dimensional setup.
