cze Funkce desaturáz a elongáz v metabolismu lipidů u diabetiků typu 2 Tato studie se zabývá hodnocením aktivity D5, D6, D9 desaturázy a elongázy ve všech třídách lipidů u diabetiků a kontrolní skupiny. Je zde zpracována závislost jednotlivých sledovaných enzymů na obsahu glykovaného hemoglobinu. V neposlední řadě se tato studie věnuje statistickému zhodnocení naměřených dat mastných kyselin a dostupných parametrů z biochemického vyšetření. Ze získaných výsledků je patrné, že aktivita D9 desaturázy je obdobná u diabetiků i kontrolní skupiny. Rovněž nebyly nalezeny korelace s množstvím glykovaného hemoglobinu. Bylo zjištěno, že aktivita D6 je u diabetiků zvýšena, čehož může být využito v monitorování lipidového metabolismu diabetiků. Rovněž byla snížená hladina PUFA u diabetiků. eng Influence of enzymes desaturases and elongases on lipid metabolism by type 2 diabetes Objectives: This study evaluates the activity of D5, D6, D9 desaturases and elongases in all classes of lipids in diabetic patients and control group. We processed dependency of individual monitored enzymes on the content of glycated hemoglobin. Finally, the study includes statistical evaluation of measured values of fatty acids and available biochemical markers. Methods: Lipid extracts from deproteinated plasma samples were separated by preparative thin-layer chromatography into five fractions (phospholipids, diacylglycerols, free fatty acids, triglycerides and cholesterol esters). All fractions were isolated and after the addition of internal standard (cis-13,16,19-docosatrienoic acid, 10 µg / ml) hydrolyzed and converted to corresponding methyl esters of fatty acids. Thus prepared samples were analyzed using a gas chromatograph. Conclusions: From the results obtained it is evident that the activity D9 desaturase is similar in diabetic and control groups. They also found a correlation with the amount of glycated hemoglobin. It was found that the activity of D6 is increased in diabetic patients, which can be used in the monitoring of lipid metabolism diabetics. There was also a reduced level of PUFA in diabetics.
