V tomto článku je popsána selektivní jodace různých 8-substituovaných imidazo[1,2-c]pyrimidin-5(6H)-onů (ethencytosinů) za mírných reakčních podmínek. Výchozí ethencytosiny byly získány cyklizací 5-halogencytosinů s chloracetaldehydem nebo postupným Suzuki-Miyaura zkříženým kaplingem mezi 8-jodoimidazo[1,2-c]pyrimidin-5(6H)-onu 1d a příslušnou arylboritou kyselinou. Reakcí [1,2-c]pyrimidin-5(6H)-onu nebo 8-jodimidazo[1,2-c]pyrimidin-5(6H)-onu 1d s N-jodsukcinimidem v DMF byl získán příslušný čistý 3,8-dijodderivát 2d. Za kyselých nebo zásaditých podmínek docházi u této sloučeniny k přesmyku na 2,8-dijododerivát 3d, který může být následně najodován za vzniku 2,3,8-trijodderivátu 4d. Vzhledem k tomu, že poloha atomů jodu na imidazolovém kruhu nemohla být přesvědčivě určena pouze pomocí NMR spektroskopie, byla dále provedena rentgenostrukturální analýza sloučeniny 2d za účelem určení a potvrzení její předpokládané struktury. Stejný sled reakcí byl použit i pro přípravu dalších osmi ethencytosinů přičemž bylo dosaženo výborné regioselektivity a vysokých výtěžků jodovaných produktů při snadném vlastním přesmyku meziproduktů. Všechny ethencytosiny byly charakterizovány pomocí 1H, 13C a 15N NMR spektroskopie.
eng Synthesis, structure and rearrangement of iodinated imidazo[1,2-c]pyrimidine-5(6H)-ones derived from cytosine We describe mild and selective iodination of various 8-substituted imidazo[1,2-c]pyrimidine-5(6H)-ones (ethenocytosines). Starting ethenocytosines were obtained by cyclization of 5-halogenocytosines with chloroacetaldehyde or by subsequent Suzuki-Miyaura cross-coupling between 8-iodoimidazo[1,2-c]pyrimidine-5(6H)-one 1d and corresponding arylboronic acids. When imidazo[1,2-c]pyrimidine-5(6H)-one or 8-iodoimidazo[1,2-c]pyrimidine-5(6H)-one 1d was iodinated by N-iodosuccinimide (NIS) in DMF, pure 3,8-diiododerivative 2d was obtained. Under basic or acidic conditions, this molecule is subject to rearrangement into 2,8-diododerivative 3d, which can be subsequently iodinated to 2,3,8-triododerivative 4d. Since the positions of iodine atoms on the imidazole ring could not be determined convincingly from NMR spectra only, X-ray analysis of 2d was carried out with the aim of confirming the structure undoubtedly. The same sequence of reactions was applied to another eight ethenocytosines, providing excellent regioselectivity, easy rearrangement and high yields of iodinated products.
