eng Optimization of Fraction Transfer Process in On-line 2D LC-CZE and Its Influence on Peak Capacity In two-dimensional separations, transfer of the analyzed fractions between separation techniques applied in both dimensions is a crucial parameter influencing the separation power of the method. 2D liquid chromatography usually utilizes valve-based interface and the attention has to be paid on miscibility of the mobile phases. In on-line coupling of liquid chromatography with electromigration techniques, the interface should also provide proper electrical connection in addition to the compatibility of mobile phase with background electrolyte. In this work, we have developed microfluidic interface for on-line coupling of both separation techniques, which was fabricated using photolithographic technique and wet etching in borosilicate glass. The fraction transfer process was studied using finite element simulation approach considering hydrodynamic and electrokinetic flow and the results were compared with the 2D analysis of oligosaccharides derivatized by 8-aminonaphthalene-1,3,6-trisulfonic acid. The second dimension electrophoretic separations in fused-silica capillary were compared to the separations in microchip format. For both techniques, in-laboratory build laser induced fluorescence detector was used, fabricated using 3D printing technology (with slightly modified design published by Přikryl [1]) and compared with the mass spectrometric detection.
