eng Automated dual comprehensive HILICxRP RPxRP liquid chromatography on a single column We investigated possibilities of combining on-line and off-line combinations of non-correlated two-dimensional systems including a single zwitterionic polymethacrylate monolithic column operated both in the HILIC and in the RP modes, in combination with silica monolithic or core-shell alkyl bonded short columns. A monolithic zwitterionic polymethacrylate micro-column prepared in our laboratory shows dual retention mechanism, HILIC and RP. The retention on the micro-column in acetonitrile-rich mobile phase (HILIC) is almost perfectly orthogonal to the retention in the RP mode in more aqueous mobile phases. This micro-column can be used in the first dimension of two-dimensional LC in alternating reversed-phase (RP) and HILIC modes, for HILIC×RP and RP×RP comprehensive two-dimensional separations. The fast repeating reversed-phase gradients on a short silica-monolithic or core-shell column in the second dimension can be optimized independently of the actual running first-dimension gradient program. We compare various aspects of three different types of second dimension gradients on the quality of 2D separation: full in fraction (FIF), segment in fraction (SIF) and shifting gradient (SG). As the alternating HILIC and RP separations on the first-dimension zwitterionic methacrylate column are based on different retention mechanisms, the instrumental setup essentially represents directly coupled two different on-line two-dimensional systems. Combining two subsequent complementary on-line 2D (HILIC×RP and RP×RP) separations provides a fast automated approach to three-dimensional data acquisition, with improved total peak capacity with respect to either HILIC×RP or RP×RP two-dimensional separations and gives us more information about the sample. two dimensional liquid chromatography; monolithic columns; second dimension gradient; HILIC; RP
