eng HPLC Analysis of Neurotransmitters and their Metabolites using Monolithic Capillary Column with Integrated Microelectrode Detector Recently developed monolithic capillary column with embedded electrochemical detector was utilized for the separation and detection of nine neurotransmitters and their metabolites. Zwitterion sulfobetaine-based monolithic stationary phase was prepared in polyimide coated fused silica capillary with internal diameter of 320 µm after thermally initiated polymerization, leaving a space at the end of capillary for incorporation of electrochemical detector. After the formation of polymer monolith, carbon fiber with diameter of 7 µm as working electrode and silver microwire with diameter of 25 µm as reference electrode were positioned into the column. Two designs of the detector with freely inserted microelectrodes or tips of electrodes fixed by the monolithic phase itself were tested. Retention characteristics of dopamine, epinephrine, norepinephrine, serotonin, 3-methoxytyramine, normethanephrine, homovanillic acid, 5-hydroxyidole-3-acetic acid, and 3,4-dihydroxyphenylacetic acid were determined in phosphate buffer with addition of acetonitrile. Optimization of mobile phase composition and flow rate was carried out to achieve a maximum separation of analyzed compounds when using capillary column containing monolithic stationary phase and incorporated electrochemical detector. Attempts to quantification of selected neurotransmitters in urine after previous preconcentration at either SPE or boronic acid-based monolithic extraction column using developed integrated capillary device will be presented.
