It represents posttranslational modification of proteins, which is correlated to various pathological states. Glycosylation controls important phenomena in the organisms like cell communication, cell recognition, protein solubility, protection from enzymes, adhesion and proliferation phenomena. The analysis of glycosylation patterns can be challenging task and may require implementation of powerful separation techniques coupled to mass spectrometry. For unequivocal identification of glycan structures, the enzymatic cleavage of glycoproteins can be used and coupled on-line with separation techniques, especially capillary zone electrophoresis and hydrophilic interaction liquid chromatography. In present work, we have developed capillary separation system suitable for comprehensive characterization of glycans. We have evaluated the possibilities of direct transfer of oligosaccharides from microreactor and injection to electrophoretic separation system, both in capillary format. Fast electrophoretic separations compatible with mass spectrometric detection were used employing volatile buffers. Various experimental setups were tested, including selective extraction and enrichment of sample compounds. The applicability of the separation system was demonstrated on analyses of deglycosylation products of hIgG and fetuin.
