cze Izotachoforetické stanovení theaninu V tomto článku je theanin stanovován v potravinových doplňcích a sypaných čajích kapilární izotachoforézou. Nejprve je theanin konvertován na aniont reakcí s kyselinou dusitou. Poté je vzniklý aniont izotachoforeticky stanoven. Optimální podmínky pro konverzi theninu jsou: roztok 0,1 mol/l dusitanu sodného a 0,1 mol/l kyseliny octové zahřívaný ve vroucí vodní lázni. Izotachoforetický elektrolytový systém obsahoval 0,01 mol/l HCl + 0,02 mol/l β-alaninu + 0,05% hydroxyethylcelulózy (pH 3,6) jako vedoucí elektrolyt a 0,01 mol/l octan sodný jako koncový elektrolyt. Kalibrační/detekční charakteristiky jsou následující: linearita v koncentračním rozmezí 15-1000 mg/l theaninu, limit detekce ca. 5 mg/l, limit kvantifikace cca. 15 mg/l theaninu. kapilární izotachoforéza; theanin; čaj; potravinové doplňky; stanovení
eng Isotachophoretic Determination of Theanine In this article, theanine is determined in food supplements and loose-leaf tea by capillary isotachophoresis. At first, theanine is converted to an anion by the reaction with nitrous acid. Then, formed anion is determined isotachophoretically. The optimal conditions for the conversion of theanine are a solution 0.1 mol/L sodium nitrite and 0.1 mol/L acetic acid heated in a boiling water bath. Isotachophoretic electrolyte system comprised 0.01 mol/L HCl + 0.02 mol/L β alanine + 0.05 % hydroxyethylcellulose (pH 3.6) as the leading electrolyte and 0.01 mol/L sodium acetate as the terminating electrolyte. The calibration/detection characteristics are as follows: linearity over the concentration range of 15-1000 mg/L theanine, limit of detection ca. 5 mg/L, limit of quantification of ca. 15 mg/L theanine.
