eng Uncommon Setup in On-line Comprehensive Two-dimensional LC Allowing Fraction Manipulation The comprehensive two-dimensional liquid chromatography separation with on-line coupling of separation dimensions was one of the most intensively studied and developed analytical techniques in recent years. Where the unidimensional LC lacks resolving power, the two-dimensional LC can benefit from the combination of orthogonal separation modes controlled by the different retention mechanisms such as reversed-phase (RP) mode and hydrophilic interaction liquid chromatography (HILIC). The application of single column with dual retention mechanism based on the composition of mobile phase can provide further increase in overall peak capacity, especially when alternating RP and HILIC separation in the first dimension is used. Using such orthogonal systems however rise the compatibility issues of the mobile phases and dilution of the sample components during the fraction transfer process. In the present work, we have therefore developed a setup for comprehensive two-dimensional LC, which allowed us to manipulate with the sample fractions after its collection and prior to the analysis in the second separation dimension. The effluent from the first dimension is collected within the interface and it is available for solvent-replacement, derivatization, or, the sample zone can be pre-concentrated and consecutively transferred into the second dimension with a narrower volume. The developed approach is an alternative to the recently introduced techniques of stationary-phase-assisted modulation and/or active solvent modulation. We have tested the setup using mixtures of porphyrine-type colorants and polyphenolic compounds, which typically presented in plants, plant extracts and food products.
