cze Vliv mobilní fáze na retenci v kapalinové chromatografii s reverzními fázemi a v chromatografii hydrofilních interakcí Cílem vysokoúčinné kapalinové chromatografie (HPLC), stejně jako jiných separačních technik v analytické chemii, je rozdělení a stanovení jednotlivých složek vzorku. Rozlišení a kvalita separace v kapalinové chromatografii je závislá na retenci látek, separační selektivitě a účinnosti chromatografické kolony. Vývoj účinných chromatografických kolon zaznamenal v posledních dvou desetiletích pozoruhodný úspěch. Nové účinné typy stacionárních fází, jako jsou kolony s částicemi menšími než 2 µm, povrchově-porézní částice, monolitické stacionární fáze nebo kolony s polem mikro sloupců, zpochybňují správnou definici objemů stacionární a mobilní fáze, mezi které je látka rozdělována během průchodu chromatografickou kolonou. Přijetí konvence fázového objemu lze zmírnit problémy způsobené možnou kombinací adsorpčního a rozdělovacího mechanismu. eng Mobile phase effects in reversed-phase and hydrophilic interaction liquid chromatography The ultimate target of High Performance Liquid Chromatography (HPLC), like of other analytical separation techniques, is the resolution and determination of sample components. The retention, the separation selectivity and the chromatographic efficiency control the resolution and the separation quality in liquid chromatography (LC). The development of efficient chromatographic columns has experienced remarkable success in the last two decades. Some of the modern types of efficient stationary phases, including the sub-2µm-particle, core-shell, monolithic and micropillar-array columns, challenge the correct definition of the volumes of the stationary and the mobile phase, between which the solute distribute during the migration along the column. Adopting a phase volume convention can mitigate the problems due to possible mixing of the adsorption and partition mechanism and to the selective preferential adsorption of mobile phase components in the stationary phase. liquid chromatography; reverse phase; Hydrophilic Interaction Liquid Chromatography; core-shell column; monolithic column; dual mode retention mechanism; prediction of retention data; retention model
