eng Composition Analysis of Complex Glycosphingolipids in Patients with Pancreatic Ductal Adenocarcinoma Revealed Differences in Comparison to Surrounding Non-tumor Tissue Pancreatic ductal adenocarcinoma (PDAC) is one of the most common causes of cancer-related deaths worldwide, accounting for 90% of pancreatic cancer with an average 5-year survival rate of less than 10% and poor prognosis. Unfortunately, PDAC exhibits aggressive biology and together with its late detection results in the majority of PDAC patients having unresectable, locally advanced, or metastatic disease at the time of diagnosis, and even traditional treatments, such as chemotherapy, surgery, or radiation, are highly resistant. Currently, lipid profiling and screening of potential biomarkers appears to be an auspicious way that may allow the early detection of PDAC or other cancer types, which could increase chances for survival. In this work, we used a modified isolation procedure originally introduced by Karl-Anders Karlsson to isolate complex glycosphingolipids (GSL) from the tumor tissues of PDAC patients and surrounding normal tissues as a control. Isolated and purified GSL were subsequently characterized using a combination of thin-layer chromatography, chemical staining, carbohydrate-recognizing ligand binding assay, and liquid chromatography coupled with electrospray mass spectrometry. Native GSL were analyzed on silica gel in HILIC mode, while GSL-derived oligosaccharides obtained by treatment with endoglycoceramidase II were analyzed using a porous graphitized carbon column. Our findings showed the major differences for GSL with five to seven sugar units and provide a platform for further studies of alterations in the composition of GSL. Glycosphingolipids; PDAC, Tissue
