Některé druhy jsou považovány za nově vznikající potravinové patogeny a mohou způsobit gastrointestinální onemocnění. Arcobacter-like bakterie byly izolovány z různých zdrojů, včetně odpadních vod, masa a mořských plodů. Detekce a identifikace Arcobacter-like species pomocí biochemických metod je problematická, a proto se častěji využívají molekulárně biologické metody. Pro detekci bakterií rodu Arcobacter z různých vzorků bylo vyvinuto mnoho PCR metod s různými primery a reakčními teplotními podmínkami. Metody PCR jsou nejčastěji cíleny na geny 16S rRNA, 23S rRNA nebo gyrA. Hlavním cílem této studie je aplikace čtyř multiplex PCR metod na sbírkové kmeny Arcobacter-like species a na jejich izoláty získané z reálných vzorků. Nejprve byla použita metoda podle Houf et al. (2000). Jedná se o rychlou, velmi spolehlivou a snadnou metodu. Metoda umožňuje identifikovat čtyři druhy těchto bakterií, což je vzhledem k objevu nových druhů velká nevýhoda. Další metoda použita pro identifikaci arkobakterů je metoda publikovaná v italské studii Daniely Pentimalli et al. (2009). Metodika mPCR popsaná v této studii je navržena k identifikaci čtyř druhů rodu Arcobacter (Arcobacter butzleri, Arcobacter cryaerophilus, Arcobacter skirrowii a Arcobacter cibarius) izolovaných především ze vzorků kuřecího masa. Metoda dle Khan et al. (2017) byla vyvinuta v Kanadě v posledních letech. Jedná se o metodu, která umožňuje identifikaci šesti druhů bakterií rodu Arcobacter. Metoda umožňuje identifikovat tradiční druhy A. butzleri, A. cryaerophilus, A. skirrowii, a A. cibarius, ale také relativně nedávno klasifikovaný druh Arcobacter, a to konkrétně A. faecis a A. lanthieri. Metoda byla vyvinuta k detekci těchto druhů právě díky jejich vzájemné genetické podobnosti. Various species are considered emerging food pathogens and can cause human gastrointestinal disease. Arcobacter-like bacteria have been isolated from a variety of sources, including wastewater, meat, and seafood. Detection and identification of Arcobacter-like species by biochemical methods are problematic, and therefore, molecular biological methods are used. Many mPCR methods have been developed for the detection of bacteria of the genus Arcobacter for different samples, with different primers and reaction temperature conditions. PCR methods are most often targeted at 16S rRNA, 23S rRNA or gyrA genes. The aim of this study was to apply four multiple PCR methods to collection strains of Arcobacter-like microorganisms and to their isolates obtained from real samples. The first was according to the method used by Houf et al. (2000). It is a fast, very reliable, and easy method to perform. However, due to the discovery of new species of these bacteria, the identification of only four species is a great disadvantage. Another method used to identify arcobacteria is the method according to the Italian study by Pentimalli et al. (2009). The mPCR methodology described in this study is designed to identify four species of the genus Arcobacter (Arcobacter butzleri, Arcobacter cryaerophilus, Arcobacter skirrowii and Arcobacter cibarius) isolated mainly from chicken meat samples. One of the methods to identify Arcobacteria in recent years was developed in Canada by Khan et al. (2017) This is a method that enables the identification of six species of bacteria of the genus Arcobacter. The method makes it possible to identify the traditional species A. butzleri, A. cryaerophilus, A. skirrowii, and A. cibarius, but also the relatively recently classified species of Arcobacter, namely A. faecis and A. lanthieri.
