eng Characterization of HILIC Stationary Phases and its Application in the Development of Gradient Separations for Metabolic Studies The hydrophilic interaction liquid chromatographic mehotd for analysis of glutathione and its related compounds has been developed. The stationary phases were selected among typical HILIC phases based on the adsorption model of aqueous layer on the surface, which was characterized using average number of water layer molecules and polar retention range determined using uracil and toluene. Stationary phases were also characterized using retention of nucleic bases and nucleosides. The retention of glutathione and related compounds was evaluated using linear and non-linear retention models, and final proposed gradient conditions were applied on the analyses of cell cultures of different densities lysed with ice-cold metaphosphoric acid. The matrix effects using mass spectrometric detection were addressed as well for the quantitative analysis of glutathione and related compounds.
