Zařízení nenese pouze základní analytické parametry, ale zároveň vykazuje vyšší citlivost, specifitu a je možná automatizace. Cílem bylo detegovat patologickou formu Abeta peptidů nebo Tau protein v CSF. Postup je založen na přípravě vysoce účinného imunosorbentu založeného na imobilizaci specifických protilátek na superparamagnetické mikro/nano částice pro aplikaci do magneticky aktivního mikroprůtokového zařízení. To vše bez negativního vlivu na funkci protilátek schopných vychytat antigen. Díky vysokému specifickému povrchu a tenké difuzní vrstvě je dosaženo vysoké kapacity nosiče a rychlé afinitní reakce. Dalším cílem je vývoj integrovaného mikrofluidního systému pro plně automatické vychytávání a charatkterizaci CTC buněk. Díky patentovanému systému Ephesia budou buňky vychytány a zároveň organizovány do jedné vrstvy umožňující automatickou in-situ imunofenotypizaci. eng	Lab-on-chip analytical system with integrated self-assembled magnetic nanoparticles for efficient capturing of biomolecules or cells from the complex biological material	We have designed and developed microfluidic multiplexed devices for rapid detection of Alzheimer disease markers. Device provides not only basic analytical parameters but also merits as increased sensitivity and specificity, possibility to automate. The goal was to detect pathological form of Abeta peptides or Tau protein in CSF. The main approach was to prepare the highly efficient immunosorbent based on immobilization of specific antibody to the superparamagnetic microspheres for magnetically active microfluidic device without adversely affecting the antibody function to capture antigen. It combines a high loading capacity and fast affinity reaction, thanks to a high surface to volume ratio and extremely thin diffusion layer. We attempt to develop an integrated microfluidic system for fully automated capture, display and typing of CTC in blood. Thanks to patented Ephesia system, cells will be captured and organized into a regular array allowing automated in-situ immunophenotyping.
