Pro vývoj metody k detekci agalaktosylované formy modelového glykoproteinu IgG, která je úzce spjatá s autoimunitními onemocněními, jsme se rozhodli kombinovat specifitu enzymových a lektinových magnetických reaktorů s vysokou citlivostí MS analýzy. ?Specifický? peptid IgG je nanopeptid odvozený z Fc fragmentu s konzervovanou N-glykosylací v pozici Asn 297. Vzhledem k velké heterogenitě aminokyselinové sekvence a přítomnosti dalších čtyř potenciálních glykozylačních míst v hypervariabilních oblastech Fab fragmentu, bylo nutné Fc fragmenty po roštěpení IgG molekuly pomocí papainového reaktoru na směs Fc a Fab fragmentů purifikovat afinitní chromatografií s proteinem A. Vysoce aktivní enzymové reaktory s imobilizovaným TPCK-trypsinem, neuraminidázou, beta-D-galaktosidázou a lektinové reaktory s Con A, GSL-I, GSL-II byly použity pro přípravu a isolaci různých glykoforem IgG specifického peptidu za podmínek vhodných pro přímou MS analýzu. Byly získány tři glykoformy tohoto specifického peptidu s experimentální MH+ 4 017 (teoreticky 4 021), 3 869 (teoreticky 3 875) a 3 711 (teoreticky 3 730). eng Enzyme and Lectin Magnetic Reactors for Analysis of Glycoproteins by Signature Peptide Approach For qualitative identification and characterization of proteins with high heterogeneity in post-translational modification and amino acid composition the signature peptides approach is preferable to peptide mapping technique. In order to develop improved method for determination of agalactosylated form of model glycoprotein immunoglobulin G which is closely associated with autoimmune diseases, we decided to combine the high specificity of enzyme and lectin magnetic reactors with sensitivity of MS analysis. Due to the high variability of amino-acid compositions and the present of additional four potential glycosylation sites in the hypervariable regions of Fab fragment, IgG molecules had to be splitted using papain reactor and obtained Fc fragments were after purified by affinity chromatography. The signature peptides of IgG are derived from Fc-fragments with conserved N-glycosylation site at the position Asn297. Highly active enzyme reactors with immobilized TPCK-trypsin, neuraminidase, D-galactosidase and lectin reactors with Con A, GSL-I, GSL-II were utilized for preparation and isolation of various glycoforms of IgG signature peptides under conditions compatible for direct MS analysis.
