Magnetické partikule představují univerzální systém s řadou výhod. Jednotlivé proteiny nebo purifikované komplexní proteiny mohou být analyzovány pomocí MS. Složky mohou být separovány (SDS-PAGE, 2D, HPLC, CZE) a následně štěpeny, nebo může být původní komplexní protein naštěpen nejdříve. Směs peptidů, které jsou výsledkem štěpení bez předešlé spearace jsou více komplexní a v některých případech mohou i nízké koncentrace kontaminantů nebo nespecifických peptidů a fragmentů komplikovat identifikaci jednotlivých proteinů. Afinitní chromatografie může být využita pro purifikaci a izolaci specifických peptidů z komplexního biologického vzorku a umožňuje jednodušší identifikaci při MS analýze. V této práci byl jako afinitní ligand zvolen anhydrotrypsin pro specifické vychytání tryptických peptidů. eng Bioaffinity microscale magnetic reactor for improved MS analysis of entire protein complex One of the modern trends in biochemistry, especially in protein and peptide analysis, is focused on the utilization of magnetic separation technique for various steps of proteomic experiment such as protein fragmentation, separation, isolation/purification and pre-concentration of peptides before final identification by mass spectrometry. Magnetic particles provide universal system with additional convenience. The individual proteins or purified protein complex can be determined by MS. Components can be separated (SDS-PAGE, 2D, HPLC, CZE) and subsequently digested, or the entire protein complex can be digested at first. The mixture of peptides that results from digestion without prior separation is more complex and in such a case even low level of contaminant as non-specific peptides and fragments can complicate identification of individual proteins. Affinity chromatography could be used for purification and isolation of specific peptides from more complex biological sample and enables simple MS identification. In this work affinity ligand anhydrotrypsin was selected as suitable affinity ligand for trapping of specific tryptic peptides.
