eng Magnetic immunospecific carrier for effective capturing of bacterial phosphoproteins suitable for implementation into microfluidic device The aim of this study was preparation of specific immunosorbent for study of bacterial phosphoproteins. Particles with different terminal function groups were tested for oriented and/or non-oriented immobilization of mono-specific anti-phospho-Tyrosine and/or combined anti-phospho-Tyrosine/Serine/Threonine antibodies. Efficiency of immobilization was verified by spectrophotometric measurement of the antibody solution before and after the immobilization, or eventually by SDS-PAGE analysis of these fractions. Binding specificity of prepared immunosorbents was checked using defined eukaryotic protein and peptide standards containing phosphorylations before application on bacterial cytosol fraction of Francisella tularensis. Phosphoproteins/phosphopeptides immunospecifically captured by the antibody from the sample were consequently eluted by the changing of pH (e. g. using 0.05% TFA or 0.16% NH4OH) and analysed by SDS-PAGE and/or MALDI-TOF/TOF-MS. bacterial phosphoproteins; Francisella tularensis; immunoprecipitation; magnetic immunosorbent
