eng SIGNAL AMPLIFICATION OF MAGNETIC IMMUNOSENSOR FOR PROTEIN DETECTION Current trend in protein detection is to minimize the sample volume. However, it includes low protein concentration and accompanying low signal response. Therefore electrochemical immunosensors get more attentions with their sensitivity and specificity. In case of such immunosensors, the attention is given to amplification of electrochemical response. One of many possibilities of signal amplification is to use the redox mediator. Thionine (phenothiazine) is a redox dye containting two amino groups in the a-positions and often used as an electron mediator1,2. In this work, thionine as the electron mediator for signal amplification is used. The aim is to improve the electrochemical response of measurement in small sample volume by addition of thionine to the substrate solution and thus more sensitive detection is achieved. To study the effect of thionine addition to the substrate solution, the immobilized horseradish peroxidase (HRP) with hydrogen peroxide as substrate were elected as model system. Such system was employed in electrochemical immunosensor for protein ovalbumin based on immunocomplex ovalbumin – anti-ovalbumin labeld by HRP formation. biosensor; ovalbumin; thionine; electrochemical detection; signal amplification
