cze Metody použité pro analýzu fosfoproteinů bakterie Francisella tularensis Posttranslační modifikace (PTM) proteinů hrají důležitou roli v regulaci různých biologických procesů. Jednou z nejdůležitějších, nejrozšířenějších a nejlépe popsaných PTM je fosforylace. Cílem této práce bylo prokázat existenci fosforylovaných forem proteinů v proteomu intracelulárního bakteriálního patogena Francisella tularensis (FT) a provést jejich detailní charakterizaci s cílem určit místo fosforylace. K obohacení fosfoproteinů/peptidů byly použity separační postupy využívající magnetické mikro- a nanočástice s povrchem modifikovaným ionty kovů, kovovými oxidy nebo specifickými protilátkami. eng Methods used for the analysis of bacteria Francisella tularensis phosphoproteins Posttranslational modifications (PTM) play an important role in regulation of different biological processes. Protein phosphorylation is one of the most widespread and crucial PTM. The aim of this work was to prove presence of phosphorylated proteins in proteome of intracellular bacterial pathogen Francisella tularensis (FT) and carry out their detailed characterization in order to determine the phosphorylation sites. We applied separation procedures using magnetic micro- and nanoparticles with modified surfaces either by metal ions, oxides or by specific antibodies for enrichment of these proteins.
