eng Electrochemical magnetic immunosensor as a versatile device for high specific and sensitive biomarker detection The high attention is nowadays focused on the development of highly specific and sensitive devices for detection of tumor markers and potential allergens due to the necessity of early diagnosis. In addition the detection of low concentrations in complex biological material and in the smallest possible sample volume is of great importance. In view of protein character of these biomarkers, combination of immunosensor with electrochemical detection and the use of the magnetic nanoparticles fulfill most requirements for versatile device for point-of-care diagnostics. In case of such immunosensors, emphasis is placed on highly sensitive response. One of many possibilities of sensitivity improvement of electrochemical immunosensor is the use the redox mediator. Thionine (phenothiazine) is a redox dye containing two amino groups in the a-positions and often used as an electron mediator. The influence of the mediator addition on the signal amplification was tested with the horseradish peroxidase (HRP) immobilized onto surface of superparamagnetic nanoparticles and hydrogen peroxide as substrate. This system simulates the final reaction of the whole immunocomplex detection. After that these findings were applied for ovalbumin detection based on immunocomplex anti-OVA/IgG-OVA? anti-OVA/IgGHRP formation. electrochemical biosensor, ovalbumin, redox mediator,
