eng Identification and characterisation of antibodies against carbonic anhydrase isoform I in intravenous immunoglobulin products Purpose/Objective: High titers of serum autoantibodies with specificity against carbonic anhydrase isoform I (CA I) were found in hematooncological patients who developed aplastic anemia type syndrome and spontaneous tumor regression after high dose therapy and autologous stem cell transplantation (HDT/ASCT) [1]. The presence of these autoantibodies had a significant survival benefit, which could be possibly used for therapeutic purposes. Anti-CA I antibodies were detected in low concentrations also in sera of healthy population. Therefore intravenous immunoglobulin (IVIG) products containing pooled polyspecific immunoglobulin G (IgG) obtained from the large number of healthy individuals plasma are a possible source of these antibodies. Material/methods: Western blot (WB) analysis was applied for verification of IVIG products (IVEGA from Imuna, Flebogamma from Grifols) reactivity with CA I. Consequently specific anti-CA I antibodies were isolated by low-pressure affinity chromatography with immobilized CA I from human erythrocytes and characterized using immunoblotting techniques. Results: Presence of anti-CA I antibodies was confirmed in both tested IVIG products by WB analysis. There were no significant differences in the reactivity with CA I antigen. Specific anti-CA I IgG fraction was successfully isolated and avidity index was defined using dot-blot affinity test. Conclusion: The results suggest that commercial IVIG products contain high concentrations of anti-CA I antibodies and could serve as source for their isolation with possible application in the treatment of some hematooncological diseases. carbonic anhydrase isoform I, intravenous immunoglobulins
