eng Influence of alcohol and pepsine presence on tramadol hydrochloride release from hydrophilic matrix tablets The aim of this work is study of influence of alcohol and pepsine on tramadol hydrochloride release from hydrophilic matrix tablets in simulated gastric fluid (SGF). In acidic medium and SGF (without alcohol and pepsin), the higher drug release rate was found for the formulations containing co-processed dry binder Disintequik™ MCC 25 in comparison with the tablets containing Prosolv® SMCC 90. Drug release rate is also influenced by viscosity grade of used MethocelTM Premium CR (K4M - higher release rate of TH). For all formulations, addition of alcohol into the acidic medium don’t influence the drug dissolution profile and drug release rate. But the differences in kinetic profiles of formulations containing Prosolv® SMCC 90 and Disintequik™ MCC 25 were found after the pepsin addition into SGF.
