eng Application of electrochemical immunosensor for detection of biologically active molecules through utilization of nanoparticle-based tags Electrochemical immunosensors are based on highly selective affinity recognition of biologically active molecules by two antibodies, one for capture from the sample and second, labeled by a suitable tag for final detection. Electrochemical techniques excel in several properties as sensitivity, simplicity, robustness, low-cost and portability. The signal-generating tags are increasingly represented by nanomaterials (quantum dots nanocrystals, metal or silica nanoparticles, graphene, etc.) which can replace commonly used labels in immunoassays, enzymes. In addition, the choice a suitable combination of nanomaterials enables simultaneous analysis of multiple substances. Here we present the development of immunosensor for analysis of ovarian cancer biomarkers HE4, CA 125 and AFP, respectively. Specific antibodies against target biomarker are labeled with quantum dots- (QDs)/gold nanoparticles-modified silica nanoparticles or QDs-modified dendrons and used for electrochemical detection. For the preparation of efficient and functional labeled antibodies, partial steps including conjugation method, set of proper dilution and optimization of affinity reaction conditions were optimized. Square wave anodic/cathodic stripping voltammetry with a compact portable electrochemical interface and disposable screen-printed three-electrode sensors allowing one drop analysis were selected for biomarkers monitoring. ovarian cancer biomarkers;electrochemical detection; quantum dots, silica nanoparticles
