eng	Simultaneous electrochemical stripping analysis of 3 ovarian cancer biomarkers using magneto-immunosensor with metal nanoparticles as antibody labels	Epithelial ovarian cancer can be considered the most lethal amongst any gynaecological malignancy mainly due to diagnosis in the late phase of the disease and the high rate of resistance to the standard chemotherapy. Nowadays, the common screening test for ovarian cancer represents the determination of cancer antigen 125 in serum which is based on enzyme-linked immunosorbent assay with optical detection. However, the specificity of this antigen to ovarian cancer is lower. Simultaneous analysis of multiple biomarkers can substantially improve early diagnosis and thus affect the survival rate of patients with this aggressive disease. The combination of commonly used magneto-immunocapture platform and electrochemical detection allows the simultaneous analysis of 3 selected biomarkers: epididymis protein (HE4), cancer antigen 125 (CA-125) and α-fetoprotein (AFP). The whole immunosensor is finally composed of primary antibodies (anti-HE4/CA-125/AFP IgG antibodies immobilized on magnetic particles) – determined antigen (HE4/CA-125/AFP) – secondary antibodies (anti-HE4/CA-125/AFP IgG antibodies labelled with SiNPs-AuNPs/QDs). The magnetic particles with covalently bound primary antibodies are one of the key components influencing the specificity and selectivity of the whole assay. They are responsible for affinity capturing and pre-enrichment of the antigen from the complex biological matrix (human serum, urine etc.). The second key assay element is the secondary antibodies labelled by 3 different types of metal nanoparticles which could be, in well-chosen combination, easily analysed by electrochemical stripping voltammetry altogether. In our work, gold nanoparticles and two core-type quantum dots (QDs) made of different metals (CdTe, PbS) and combined with silica nanoparticles were found as a suitable combination for simultaneous analysis.
