Balzamikový ocet je jedním z nejznámějších a nejoblíbenějších octů na světě a je významným zdrojem fenolických sloučenin. Kvalita balzamikových octů i obsah fenolických látek se liší v závislosti na způsobu výroby. V této práci byla vyvinuta metoda kapalinové chromatografie spojenou s MS a MS/MS detekcí pro kvantitativní stanovení obsahu fenolických sloučenin v balzamikových octech. Optimalizace parametrů MS/MS a separačních podmínek, tj. výběr kolony, složení mobilní fáze a gradientový profil, byla provedena pro 32 standardů fenolických sloučenin. Jednoduchou přípravou vzorků bylo analyzováno celkem 14 vzorků různých druhů balzamikového octa. Profil fenolických sloučenin byl doplněn semikvantitativní analýzou těkavých organických látek pomocí metod GC-MS a GC-FID. Extrakce VOCs byla provedena metodou HS-SPME, pro kterou byly nejprve optimalizovány parametry extrakční teplota, doba extrakce a objem nasyceného roztoku NaCl. Dosažené výsledky byly porovnány s již existujícími pracemi. Kyselina gallová, kyselina protokatechuová, kyselina kávová a kyselina p-kumarová byly identifikovány jako hlavní fenolové sloučeniny v analyzovaných balzamikových octech. eng Analysis of Phenolic Compounds using HPLC and GC Methods in Different Types of Balsamic Vinegars Phenolic compounds affect the properties of food products and are important antioxidants. Balsamic vinegar is one of the best known and most popular vinegars in the world and is a good source of phenolic compounds. The quality of balsamic vinegars as well as the content of phenolic substances varies depending on the production method. In the present work, we have developed liquid chromatographic method coupled with MS and MS/MS detection for quantitative determination of the content of phenolic compounds in balsamic vinegars. Optimization of MS/MS parameters and separation conditions, i.e. column selection, composition of mobile phase and gradient profile, was performed for 32 standards of phenolic compounds. A total of 14 samples of different types of balsamic vinegar were analyzed with simple sample preparation. The profile of phenolic compounds was completed by semiquantitative analysis of volatile organic compounds using GC-MS and GC-FID methods. VOCs extraction was performed by the HS-SPME method, for which the parameters of extraction temperature, extraction time and volume of saturated NaCl solution were first optimized. The achieved results were compared with already existing works. Gallic acid, protocatechuic acid, caffeic acid and p-coumaric acid have been identified as the major phenolic compounds in balsamic vinegars. Phenolic Compounds; HPLC; GC; Balsamic Vinegars
