The group of heavy metals comprises essential elements necessary for the proper function of human metabolism, but also metals that pose a risk of toxicity to the human body. Exposure to cadmium and mercury can damage the cells of kidney, liver, lung, or brain. Especially in neural cells, cadmium and mercury can induce functional and morphological changes at low concentrations. Because these two heavy metals are widespread in the environment, it is essential to study their toxicity. We aimed to characterize the biological effects of CdCl2 and HgCl2 on glutathione levels and dehydrogenase activity in the human neuroblastoma SH-SY5Y cell line. Cells were exposed to CdCl2 and HgCl2 (0-200 μM) for 6 and 24 h. In addition, changes in cell morphology after treatment with CdCl2 and HgCl2 were observed using fluorescence microscopy. We found changes in dehydrogenase activity and glutathione levels after 6 and 24 h incubation with CdCl2 and HgCl2. After 24 h, we found a transient increase in glutathione levels after incubation of cells with the lowest tested concentrations 5 µM and 1 µM of CdCl2 and HgCl2, respectively. After 24 h, decreases in dehydrogenase activity and glutathione levels were detected after incubation with both 100 μM CdCl2 and 50 μM HgCl2. Furthermore, these results were supported by fluorescence microscopy. In conclusion, our results showed that HgCl2 induced a larger impairment of neural cells compared to CdCl2.
