eng Study of acetaminophen hepatotoxicity in vitro - role of glutathione reductase inhibition. Acetaminophen (paracetamol, APAP) is one of the most used antipyretics and analgetics in present. It is a safe drug at therapeutical doses; however, the overdose may lead even to the acute liver failure. The aim of our work was to find if the glutathione reductase (GR) inhibition may contribute to the impairment of APAP treated cells. We used rat hepatocytes isolated by two step collagenase perfusion and cultivated on Petri dishes. The inhibitory effect was assessed in a solution where GR prepared from rat hepatocytes and the other reagents were present (NADPH, GSSG). We found an inhibitory effect in the presence of APAP-SG; we proved a decrease of GR activity by 30 % and 60 %, in 1.5 mM and 3.7 mM APAP-SG treated samples, respectively (compared to control). The described inhibitory effect of APAP-SG conjugate may play likely an important role in the mechanism of APAP toxicity due to deepening of the typical glutathione depletion in the hepatocytes.
