eng Chip-based bioaffinity assays: new tool for protein analysis Affinity purification is powerful separation method based on the specific interaction between immobilized ligands and target proteins. Unlike some other chromatography systems, affinity purification can be performed with modified batch methodologies and does not require a packed column. Using carriers with magnetic property overcomes many of the problems associated with the use of liquid gel slurries in high-throughput and standard laboratory applications. Magnetic particles provide universal system with additional convenience, consistency, stability, ease of handling and exceptional flexibility compared to standard chromatography resins. Implementation of the bioaffinity assays on the microchip format allows faster analysis times due to favorable reaction kinetics on the microscale format. The reduction of sample volume and high-cost reagent consumption are further important priorities. An objective of this presentation is to perform new microsystem using superparamagnetic nanoparticles as a self-organizing matrix. The versatility of new microfluidic device enables integration of different reaction steps in a microchannel. This contribution covers three types of bioaffinity assay: enzymatic assays (specific fragmentation of protein), peptide extraction technique and isolation of target molecules out of complex matrix (tryptic peptides purification). affinity chromatography; microchip; immobilized antibodies, immobilized enzyme
