eng Optimization of Off-line and Heartcutting On-line LC-MEKC Separation Employing Sweeping Preconcentration Step Two-dimensional separation methods based on combination of liquid chromatography in one dimension with electromigration separation in the second one offer highly orthogonal separations with considerably increased number of resolved compounds. In this work, a 2D method combining liquid chromatography in the first dimension with micellar electrokinetic capillary chromatography in the second dimension was applied for the separation of phenolic acids and flavone compounds. The packed and monolithic microcolumns and (poly)etylene glycol columns previously used in 2D LCxLC separations were employed in the first dimension for HPLC separation. The automated off-line and heartcutting on-line techniques for connection of LC and CE instruments were optimized and compared. In off-line setup, a CE autosampler was used as the interface providing automated fraction collection of LC effluent and injection into the second dimension MEKC separation.
